摘要:以黄芪多糖(APS)为碳源,经Cu^(2+)掺杂,一步水热法制备了铜掺杂黄芪多糖衍生碳点(Cu-ACDs)。采用UV-Vis、FL、FTIR、TEM和XPS对Cu-ACDs进行了表征。通过CCK-8法测试了Cu-ACDs对L929细胞的细胞毒性和增殖能力,通过游离血红蛋白检测评估了Cu-ACDs的溶血性能,采用自由基清除实验考察了Cu-ACDs的抗氧化活性。结果表明,Cu-ACDs为类球状粒子,平均粒径为5.14 nm,呈绿色荧光,荧光量子产率为22.4%,其在490 nm激发波长下的发射波长为560 nm。Cu-ACDs质量浓度为200 mg/L时,L929细胞保持良好活力,在第3 d和第5 d时,细胞明显增殖。Cu-ACDs质量浓度在50~800 mg/L时,溶血率均<5%。与APS-Cu复合物和未掺杂铜黄芪多糖衍生碳点(ACDs)相比,Cu-ACDs不仅对金黄色葡萄球菌、大肠杆菌的抑菌圈增大,而且对黄曲霉菌和黑曲霉菌也表现优异的抑制活性,上述4种菌的最小抑菌质量浓度分别为60、100、125和40 mg/L;Cu-ACDs对1,1-二苯基-2-三硝基苯肼自由基、2,2'-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐阳离子自由基和·OH的半数清除质量浓度分别为48.6、6.4和3.8 mg/L。
Cu-doped carbon dots(Cu-ACDs)was prepared by one-step hydrothermal method using natural Astragalus polysaccharides as carbon source and Cu^(2+)as doping agent,and characterized by UV-Vis,FL,FTIR,TEM and XPS.The Cu-ACDs obtained was then analyzed by CCK-8 method for its cytotoxicity and proliferation promotion capacity on L929 cells,by free hemoglobin detection method for its hemolysis performance,and by free radical scavenging assay for its antioxidant activity.The results showed that Cu-ACDs displayed a morphology of globoidal particle with an average particle size of 5.14 nm,and exhibited green fluorescence,with a fluorescence quantum yield of 22.4%and an emission wavelength of 560 nm at 490 nm excitation wavelength.L929 cells co-cultured with mass fraction 200 mg/L Cu-ACDs demonstrated excellent vitality and significant proliferation on the 3rd and 5th day.The hemolysis rate of Cu-ACDs for rabbit blood in the mass concentration of 50~800 mg/L was lower than 5%.Compared with APS-Cu complexes and undoped Astragalus polysaccharide derived carbon dots(ACDs),Cu-ACDs improved the antibacterial activity against Staphylococcus aureus and Escherichia coli,also exhibited better inhibitory activity against Aspergillus flavus and Aspergillus niger.The minimum inhibitory mass concentration of Cu-ACDs for the above strains were 60,100,125 and 40 mg/L,respectively.Meanwhile,the median inhibition mass concentration of Cu-ACDs against 1,1-diphenyl-2-trinitrophenylhydrazine radicals,2,2'-diazide(3-ethylbenzothiazolin-6-sulfonic acid)diamiammonium cation radicals and?OH were 48.6,6.4 and 3.8 mg/L.
